To prepare the transverse section of the stem, I used a scalpel to cut a thin section of the stem. The section was then immersed in water to prevent it from drying out, and a cover slip was placed over the section to prepare it for examination under the microscope.

Once the slide was placed on the stage of the microscope, I first turned on the light source and used the low power objective lens to locate the section. I looked through the eyepiece and carefully adjusted the focus using the coarse focus knob. Once the section was in view, I switched to a higher power objective lens to obtain a more detailed view, and finely adjusted the focus with the fine focus knob.
