To make a section of the stem, I used a scalpel or a backed blade to carefully cut the stem into thin slices. The sections were then prepared by placing them onto a slide, and I added a few drops of water. I also covered it with a cover slip to avoid contact with air.

I started by ensuring the light source was on and used the low power objective first. I looked through the eyepiece and moved the slide until the section was in view. I then adjusted to a higher objective for more detail, using the coarse focus knob initially followed by the fine focus knob.
